That’s probably why Spike and Twilight don’t call each other brother/sister or son/mother. They feel that they are a mixture between the two. But they’ll always know they are each other’s friends.
True, it’s never been confirmed that they are mother/son or sister/brother, but I imagine Twilight as an older sister who’s responsible for Spike. She was only a kid when she hatched him, so someone else (Celestia, Twilight’s parents) must have raised him until Twilight was old enough to take care of him. Though I can see why Twilight is considered in a way a mother to Spike.

Twilight may not be his biological mother, but she’s his adoptive mother.
They don’t reffer to each other as ‘mommy’ or ‘sister’
But Twilight hatched and takes care of Spike, doing things like bathing him and telling him to stop eating ice cream and taking him to the doctor, so she is as close to a mother as he has.
Spike and Twilight have a more ‘brother-sister’ sort of relationship.
I’ve seen that idea as well, but in Winter Wrap Up, Spike says to Twilight “You’re not mommy” and she doesn’t seem upset or shocked.
