The word “unicorn” appears in the King James translation of the Bible. It came from the Latin word unicornus found in the Roman Vulgate Bible, which in turn was the translation of the Greek word monocerous found in the Septuagint, a Greek translation of the Jewish Scriptures. The Greek word was the translation of the Hebrew word re’em, which apparently was some kind of fantastic beast in Jewish folklore.
 
No one’s really sure what a re’em was supposed to be. It might’ve been based on the aurochs, the now-extinct wild cattle, or possibly some kind of oryx or other wild antelope. The Biblical context indicates that whatever it was, it had a horn or horns, was very big, extremely strong, and completely wild and untameable.
 
This latter part was the origin of the whole “virgins and unicorns” thing. Medieval folk believed that only a maiden pure of heart, i.e. a virgin, could approach such a wild and shy creature. Anyone else would surely be killed by the fierce beast.

Technically speaking it’s more an issue of translation, the original text was written in a language that has a lot of compound or otherwise nonspecific words, so terms that can be translated as unicorn could also mean things like rhinos.

Yeah, that’s a mistranslation on par with “suffer not the witch to live.” In the original Hebrew, that one’s referring to aurochs, a.k.a. wild cattle.
