Some fans actually used to speculate if they were related, I think that’s what this is spoofing.

People are really grasping for straws here. Rainbow Dash was merely comforting a filly she barely knew that had been through a horrible ordeal. She was probably barely listening to that adrenaline-panic-filled babbling Scootaloo was spouting at that moment…

Scootaloo specifically said (quoting from the transcript here): “I’m so, so sorry, Rainbow Dash! I just wanted you to hang out with me and see how cool I was so you’d take me under your wing, teach me everything you know, and become like my big sister!” [my bold].
 
Dash’s response was to take Scoots under her wing, both metaphorically and literally.
 
Also, this at the end of the episode:
 
Sweetie Belle: I call sister teams! Last herd to make it to the falls is a moldy carrot!  
Rarity: Ugh, if you insist. [giggles] It is so on!  
Rainbow Dash: They think they can beat the two of us?
 
It’s pretty clear from context that Dash is treating Scootaloo as a sister, even if she hasn’t legally adopted her.

I like that they’re not related. It adds variety, and like AJ adopting Pinkie, it reinforces that your friends and family are the people you care deeply about, not simply the people whose DNA you share.
 
Dash didn’t have to adopt Scootaloo as her little sister. The fact that she did, and that that’s obviously quite important to her, makes the relationship stronger and shows Dash as a better and more mature pony than she used to be.
