My guess is that Poison joke effects unique things about individual ponies.
Fluttershy’s soft voice is what poison joke effects.
Like Applejack being big and strong being small and weak.
Zecora made a potion with it, so I think it was tampered with to get the desired affect.
Okay, let me get this straight: Is the Poison Joke supposed to be randomized or is is specific for each individual pony?
‘cause if it’s the former, I’m calling bullshit on how it managed to have the exact same affliction for her twice in a row.
In order to replace Big Mac’s voice in the The Ponytones, Zecora made a potion out of poisonjoke to turn Fluttershy back into Flutterguy. Hence the ‘Flutterguy’ tag.
