I think the joke is that everypony is crying tears of joy for the couple not that they are jealous.
Contrary to what seems to be popular thought, not every single mare wants Big Mac. A lot do of course but you get the point
The joke is that Rainbow isn’t into this sappy stuff.

So how does this count as a lesbian joke? Because like I said a Lesbian can cry at weddings. Any man or woman can. Just not everyone does. She may just think Rainbow Dash isn’t a cryer.

Pathetic Failure
As much as I like to headcanon Rainbow as gay (or bi), I doubt this is a subtle joke about it. From my perspective, Rainbow is simply looking at Fluttershy and is wondering if she is crying or yawning.
 
Also, Rarity is too busy trying to catch the bouquet to cry. As the old legend goes: Whoever catches it will be the next to be married.

They all crying because, in Fleetfoot’s mind, Big Mac is the stallion that every mare would want to marry. There is no other reason than RD’s being gay that she’s not crying.

It doesn’t mean she’s a lesbian. But it would be intensly out of character for Rainbow Dash to cry over anything like a wedding.
