It’s perfectly understandable that neither Spike nor Twilight ever really though about it and stick to “best friend and assistant” theme. That said, you can’t describe their relationship in “only this” or “only that” terms. No matter how you describe it, at the end he is very attached to Twilight and she is his only caregiver.

Genius, we’re not saying she’s ACTUALLY his mother, just that they have a relationship that’s halfway between siblings and mother and son because she’s his guardian and takes care of him.

What are you talking about? Of course they do! And they give them baths and tuck them in and keep them from eating too much ice cream and take their comics when they stay up too late and… um… They don’t, do they?

Because everyone embraces their secretaries like in the pic. Or takes them to the doctor when they’re looking weird. Or dries their tears and comforts them when they are sad.
 
Spike is like a younger-brother/son and life companion, one that assists and helps Twilight because he’s become competent and good at it.
