Considering in this case all it needs to work on is the trigger, it more than likely already did.

That definitely isn’t an exit hole, so aside from the artist’s intent on the small graphic intending to be an exit hole, the next assumption is that somepony on our end shot first, but the target was layered and against the proper face of the silhouette, yielding way for Twi to have the advantage. The only reason we don’t see the second shot against us is because she is either correcting us on the right end of the target, or she aimed well and very well at our head, the falling target and emerging Twilight being the last thing we see.
 
Overanalysis aside, that is a really good Weaver hold considering hooves.

Judging by the bullet holes nature, somepony shot through the target at Twilight first.
 
Although, how the target falls does indicate this was not shot from behind, so my first assumption that Twilight had shot through the door at the viewer is now false.
 
Still…
