It’s a pretty easy thing to miss, nice catch.
I’ve only watched the full episode once, too, and had to look up the scene to get the line right, but the detail of them being (fraternal) twins stuck with me.
I’ve only seen that episode once, and I can’t remember every line. Interesting.

But there’s two kinds of twins: identical/monozygotic and fraternal/dizygotic.
All twins (that I can remember off the top of my head, at any rate) in the show are in the latter category, which was kind of the point of those first two comments.
Hence, my earlier remark.
This. If memory serves, the writer of that episode explained that it depends on what your definition of “twin” means, but Pinkie and Marble would fit his.
I never heard the show call Pinkie a “twin.” (Granted pretty much almost every pony looks the same, but with different eyes, hair, colors.)
Oddly, the SNES Yoshi’s Island game intro refered to the Mario Brothers as “twins,” but that was oddly removed from the GBA remake. Can’t have anything set-in-stone for the Mario series, I guess.

However, let’s not forget Equestria is a magical place. Quite literally. The rate of twins appears to be rather high in general. Technically, even Pinkie Pie is a twin!
Identical twins are produced by the zygote splitting in half very early on, so the two have the exact same DNA.
So yes, the only way to get twins of the opposite sexes (let alone different races) two egg cells need to be fertilized from the get-go.
I hear identical twins can’t be different genders, so they must be fraternal twins. (Especially, considering they’re different pony races.)
