*1-*The sisters are immortal because they’re alicorns (unlikely due McCarthy’s comment on Twilight not outliving her friends. And its unlikely they’d make the Mane5 alicorns as well).
 
*2-*The sisters are immortal because of the sun/moon thing (Twi/Cadance isn’t immortal).
 
*3-*The sisters are not immortal, just long-lived (For example, if we were to assume they are around 3000-4000 years old, that could be the equivalent of a 30-40 year old pony).  
But they’d still be mortals.
 
*4-*The sisters have normal lifespans, no longer than any other pony, but have been using the Age spell to keep themselves young for a specific reason (finding a successor to take over their tasks, guiding the new elements of harmony to adjust to their roles, etc).
 
I guess there could be more options, but those sound as the most likely ones, I guess.

Yes, I know. Im just saying that assuming they’re immortal (and not just long-lived) is pure headcanon, since living over 1000 years doesn’t automatically imply immortality.  
My post was mostly directed to those who still believe the sisters to be some kind of gods, which has no basis. Heck, Magic Duel even opened the possibility that the sisters may have lived that long due to the age spell, and not because of an actual long lifespan.
