The joke is that Rainbow Dash is challenging Big Mac to a game of soccer as a sort of test of character, like she did to Twilight in Equestria Girls, but Big Mac misinterprets it as her saying she wants to have sex with him to see if he’s good enough at sex for Fluttershy.
