I just figured it out:
Fill both of her bars as far as you can, when the transparent bar is flashing, you have to turn the pump buttons to off then CONSTANTLY switch between fast and slow for the vibrator. It’ll fill VERY slowly, but it DOES get the job done.
But RD’s climax bar fills too fast and her milk bar drops too fast. What’s the proper settings to stop that from happening?

Actually, that is not quite true. You don’t need to use specific characters to unlock the others, with the exception of Twi. The unlocks are as follows:
Applejack - Use only the pump.
Pinkie Pie - Use only the vibrator.
Rarity - Overfill the tank.
Fluttershy - Do not use the fast settings.
Twilight - Achieve orgasm with the rest of the mane 6.
Derpy - Achieve orgasm, but cum nothing (this will show up as a broken heart in the speech bubble).
