OK, here’s a guide for starters: if you know Kinnikuman, jump to the second paragraph.
 
Kinnikuman is a Japanese wrestling/humor/superhero manga by the team Yudetamago (literally, Boiled Egg). The animated version of the adventures of the titular character was a considerable hit not nonly in Japan, but in other parts of Asia and Europe as well. In the States its characters got famous as a collection of rubber figures made in the 80s, called M.U.S.C.L.E. One of the main characteristics of this manga is that the characters were created by fans, who would post their drawings to the authors. They were more than happy to oblige their requests. Still are: the series is still running, and strongly enough. About the two characters, Pinkie turned into Ramenman, a Chinese hero. The guy with the Tia-like appearance is Kinnikuman himself.
 
And about the joke itself… It’s about what’s beyond the 4th wall and otherwise, I think it doesn’t make actual sense (but man it’s funny and a treat to the fans).
