The way I see it, she felt she had done nothing important with her title as princess, not that she had never done anything important. At the end though she realized she’s a princess for what she’s been doing all along (while not princess), so everything she’s been doing this season have actually been princess accomplishments, even if they’re not political in nature like what the other princesses seem to do.
First there is a mountain, then there is no mountain, then there is.
The song they sang to her felt like it was more like rubbing it in her face.
I’m pretty sure that up to this moment, she’s learned magic of friendship, not spread.
The funny thing is, Twilight was lamenting how she never did anything important at the start of the episode, being relagated to smiling and waving at official meetings.
So, Twilight at the start: “I never do anything exceptionel, I just keep doing what I’ve always done, nothing have changed”
At the end: “I’m going to keep doing what I’ve always done, nothing will change, this is the role I have chosen for myself”.
Doesn’t look like Luna and Celestia are mocking her, they just appreciate the irony of Twilight’s statement.
I think you mixed it up.

Their rule-role:
Princess of Equestria
Princess of Equestria

I guess Equestria is triarchy now.
Besides saving the entire world along with her friends time and time again, right?
And that’s unreasonable and mockable because….?
It’s not like she’s been doing bad things, I don’t get what kind of eventual role people were expecting, it’s kinda in the title of the show, and her title fits perfectly alongside the other princesses and just corroborates with the themes and messages of the show.
So her purpose is what she’s been doing…that sounds completely reasonable, not mockable.
Actually Derpy is backup princess, but she was unavailable at the time.
S02 Cadance: spread Magic of Love, no castle.
Current Twilight: spread Magic of Friendship, with own castle.
I say, Twilight is getting better than S2 Cadance here.
Her “self-affirmed” royal purpose is to carry on as per usual.

Well, let’s look at the facts.
Princess Celestia raises and lowers the sun.
Princess Luna does the same for the moon.
Princess Cadance rules an entire Crystal Empire.
And Twilight…sadly, doesn’t do anything that impressive.
