Technically she’s not a background pony. She’s a secondary character (Not a main but more than an extra) Though I imagine even pony Cherilee has to deal with tons of stress. The difference is most likely the society around them. Pony society focuses heavily on happiness, joy, and unity. Human society..well…

she look so tired and annoyed, one of the few thing I didn’t like of MLP:EG, Cheerilee is one of my faves bgp, it was sad looking at her like that :(

Human world Cheerilee seems to be older than pony world Cheerilee, so she’s had longer to become bitter and disillusioned.
 
Pony Cheerilee also deal with younger students, so she probably puts more effort into presenting a positive attitude.
