Background Pony #43E4
Optimus Prime is 38 ft / 11.58 meters tall if he is from the IDW Comics. A little wrong here, but the Twilight Sparkle is not much bigger than a regular MLP G4 pony. A little unclear, but it looks like she’s human size. Also, it is common for Optimus Prime and Megatron to be larger than the other transformers across the size rules for hands on cartoonists. He can be far bigger than stated.
