Autism is the instinctive ability to recognize social constructs. That is all.
It can manifest in different behaviors and levels of functionality that may/may not involve socially acceptable behavior (“rude” behavior), but includes a learning curve to established social constructs.

You tell a young Autistic child to “always tell the truth”, but when they call somebody “fat”, you tell them not to say things like that and to “be nice” instead. In absolute reality, telling someone fat is not morally wrong or incorrect behavior; perceiving it as “rude” is a social construct. The child instinctively sees this even when they don’t understand it.
The contradiction creates a fearful paradox that often results in withdrawn and reclusive behavior, pull towards other Autistics for company, interests that seem eccentric, and/or seemingly unruly behavior at ignoring or struggling to adapt.

PINKIE: Lack of personal space, difficulty understanding certain “normal” behaviors, believes everything can be solved with cupcakes and parties, shows overt friendliness even when unwarranted, erratic attention span, misunderstands “emergencies”, does not react appropriately to many situations (giggles and makes faces in horrifying darkness in a wild forest, but screams wildly at every opportunity in Nightmare Nights in pretend danger), doesn’t always acknowledge proper manners, possesses an uncanny and extensive filing system
Gives out way too many hugs

MAUD: Definitely Asperger’s. Monotone, feels several emotions but doesn’t manifest them in expressions most can understand, super chill, extremely straightforward, ignores social greetings, obsession with rocks, often requires others’ initiative, enjoys others’company at nearly any cost even against her wishes
Very rarely gives hugs, and almost never returns them

LIMESTONE: Reacts to social interactions, constructs, and emotional behavior with anger, frustration, and aggression as a defensive mechanism, overtly focused workaholic, and very pragmatic
Hates hugging altogether, misunderstands and ignores the point of the gesture

MARBLE: Easily fearful and embarrassed in practically all social situations, similiar difficulty in expression, overtly submissive to avoid confrontation, extremely reclusive and withdrawn except to her sisters
Wants affection, but always finds hugging awkward… well everything seems awkward

It’s not a disease, it’s just different. I’m not Autistic, but I prefer the company of Autistic friends because I recognize social constructs and outright reject them on purpose, and it’s easier to enjoy company of others without that. Autistic people in turn seem to likewise enjoy my company because they feel relaxed without fearfully being upright at saying/doing something socially wrong. Not to mention teaching them it’s society that’s broken, not them.

Dash canonically can’t outrun Murder Mode Pinkie, though, since the laws of physics are more like suggestions to her.

It is a disorder, a spectrum one.

Each Pie sister is a different aspect on the Autistic spectrum.
It can be difficult for most people to tell because they don’t realize Autism is more like personality traits, not a group of disorders.

The race would have to be tailored in such a way as to make Pinkie not overtly speedier than RD, otherwise that just sounds dangerously close to Mary Sue territory.

Making Pinkie an Earth Pony and Fluttershy a Pegasus afraid of heights was the right call, even if our pony goddess feels like she betrayed Pinkie.

imagine if Pinkie was a Pegasus there would be an episode where Rainbow gets TOO Prideful, belittles her friends and such, and Pinkie teaches her a lesson by getting serious and DESTROYS Rainbow in a race, despite of her wanting to play for fun rather for pride
